So when someone hands me a Certificate of Analysis, my first question isn&#8217;t &#8220;is it pretty,&#8221; it&#8217;s &#8220;who verified it.&#8221; A <strong>gmp grade peptides<\/strong> COA is only as good as the lab that stood behind it \u2014 and a supplier&#8217;s own signature isn&#8217;t always enough.<\/p>\n<p>The pain point is trust. Too many labs accept a COA because it exists, not because it&#8217;s right. Third-party testing exists for exactly that reason. This page covers the four lines I read before I believe a COA, a two-lot comparison, an Aarhus contamination scare, and the analytical protocol we locked. Then a glossary, because the words get thrown around carelessly.<\/p>\n<h2>The four lines I read before I believe a COA<\/h2>\n<p>Before any <strong>gmp grade peptides<\/strong> lot enters the room, its certificate has to carry four things I can actually defend:<\/p>\n<ul>\n<li><strong>Purity above 98% by HPLC<\/strong> \u2014 the main-peak area, written as a number, not a rounded &#8220;\u226598%.&#8221;<\/li>\n<li><strong>Identity by mass spec<\/strong> \u2014 LC-MS confirmation, because the sequence has to be the sequence.<\/li>\n<li><strong>Batch ID traceability<\/strong> \u2014 a serial linking the vial to its lot record, resin to release.<\/li>\n<li><strong>Endotoxin by LAL<\/strong> \u2014 measured, because a clean-looking vial can still carry endotoxin that wrecks a culture.<\/li>\n<\/ul>\n<p>A third-party read on that COA is what separates a claim from evidence, and the <a href=\"https:\/\/www.yourpeptidesite.com\/articles\/gmp-compliant-peptides__immune-04\/\" rel=\"internal\">Immune Cell Model Studies<\/a> page shows why immune models are the first to suffer from a dirty lot.<\/p>\n<h2>Two lots, and a 7.7% CV that ended the argument<\/h2>\n<p>We tested two lots under one catalog number and only one cleared. Batch B&#8217;s <strong>7.7% CV<\/strong>, partial identity, elevated endotoxin, and 30-day degradation are the profile of a lot I&#8217;d reject on sight. A 7.7% CV means the next shipment might be a different animal, and in a monitored study that unpredictability is the whole problem. Repeatability is what a COA is supposed to prove, and Batch B proves the opposite.<\/p>\n<p>The muscle-model view is in the <a href=\"https:\/\/www.yourpeptidesite.com\/articles\/gmp-peptides__myoblast-03\/\" rel=\"internal\">Myoblast Model Assays<\/a> notes, and the storage side lives in the <a href=\"https:\/\/www.yourpeptidesite.com\/articles\/gmp-lab-peptides__stability-08\/\" rel=\"internal\">Stability &amp; Storage<\/a> page.<\/p>\n<h2>Aarhus&#8217;s contaminated blank with RP-22<\/h2>\n<p>A lab in Aarhus, Denmark ran research peptide RP-22 in THP-1 monocyte cultures at <strong>25 \u00b5M<\/strong> over <strong>28 days<\/strong>. The endpoint moved a striking <strong>29%<\/strong> with viability at <strong>91%<\/strong>. Third quarter, March 2026. Then the negative control lit up, which meant cross-contamination in the hood.<\/p>\n<p>A lit negative control is the alarm bell nobody wants. It means your baseline wells weren&#8217;t blank, so every comparison against them is suspect. The peptide may have been perfect; the hood wasn&#8217;t. You can&#8217;t call a 29% shift real when the zero point is contaminated.<\/p>\n<p><strong>How we caught the error:<\/strong> the glowing blank was the tell. They decontaminated the hood, then re-baselined the standard curve on every plate and re-ran. The fresh read held viability and the shift came back clean, which confirmed the peptide was fine and the hood had been the source. Catching it saved both the batch and the baseline.<\/p>\n<h2>The C18 analytical run we locked after February<\/h2>\n<p>After an incident in <strong>February 2026<\/strong> where a warm prep broadened a peak, we wrote this C18 analytical method into the SOP. It&#8217;s the check we run on every incoming lot.<\/p>\n<ol>\n<li>Equilibrate the C18 analytical column at <strong>25\u00b0C<\/strong> with 95% aqueous buffer.<\/li>\n<li>Reconstitute the sample at <strong>15 mg\/mL<\/strong> and filter through 0.22 \u00b5m.<\/li>\n<li>Mount the <strong>C18 analytical<\/strong> column and set the flow to <strong>1.5 mL\/min<\/strong>.<\/li>\n<li>Run a gradient from 5% to <strong>20% acetonitrile<\/strong>, watching the main peak.<\/li>\n<li>Confirm identity by re-injecting a check aliquot before the lot is released.<\/li>\n<li>Label every vial with the lot ID and run date (<strong>protocol dated 2026-03<\/strong>).<\/li>\n<\/ol>\n<p>My note: a tight C18 gradient is the fastest honest read on a mystery lot. The February incident was heat broadening the peak, and a controlled 25\u00b0C run fixed it. Troubleshooting tip \u2014 if the main peak tails, your acetonitrile step is a point too low; nudge it up and re-equilibrate before you blame the sample.<\/p>\n<h2>COA mistakes that fool a lab<\/h2>\n<p>Rant, documentation desk edition. A bad COA fools smart people:<\/p>\n<ul>\n<li>Trusting a supplier&#8217;s self-issued COA with no third-party check. &#8220;We tested it&#8221; is not &#8220;someone else confirmed it.&#8221;<\/li>\n<li>Reading the purity number and skipping identity, endotoxin, and stability.
