Batch B&#8217;s <strong>6.1% CV<\/strong>, elevated endotoxin, and 30-day degradation tell me that lot was already sliding before it left the dock. High CV in a dermal study means your collagen signal moves for reasons that have nothing to do with biology. They opened at <strong>200 \u00b5M<\/strong> for a short <strong>7 days<\/strong>. The endpoint moved <strong>12%<\/strong> with viability at <strong>91%<\/strong>. Third quarter, May 2026. Then the peptide aggregated at 200 \u00b5M, so they had to drop to 50 \u00b5M and re-run.<\/p>\n<p>Aggregation at high dose is the classic trap. That read is what told them the high-dose well was never in solution, and the 50 \u00b5M re-run became the number they kept.<\/p>\n<h2>The C18 analytical run we set after March<\/h2>\n<p>After an incident in <strong>March 2026<\/strong> where a warm prep smeared a peak, we locked this C18 analytical method into the SOP. Cold, tight, repeatable.<\/p>\n<ol>\n<li>Chill the C18 analytical column to <strong>4\u00b0C<\/strong> and equilibrate with 95% aqueous buffer.<\/li>\n<li>Reconstitute the sample at <strong>20 mg\/mL<\/strong> and filter through 0.22 \u00b5m.<\/li>\n<li>Mount the <strong>C18 analytical<\/strong> column and set the flow to <strong>1.0 mL\/min<\/strong>.<\/li>\n<li>Run a shallow gradient from 2% to <strong>5% acetonitrile<\/strong>, watching the main peak.<\/li>\n<li>Confirm identity by re-injecting a check aliquot before the lot is released.<\/li>\n<li>Label every vial with the lot ID and run date (<strong>protocol dated 2026-05<\/strong>).<\/li>\n<\/ol>\n<p>My commentary: a shallow gradient on C18 is unglamorous but it separates a clean main peak from the junk that wrecks dermal assays. The March incident was heat smearing the resolution, and I haven&#8217;t trusted a warm prep since.
